Reagents and materials:
Medium
2. 0.05% trypsin containing EDTA;
3. PBSA;
4. T25;
experimental method:
1. To obtain freshly prepared CB-MNCs, separate mononuclear cells (MNCs) by density gradient separation;
2. Resuspend freshly isolated or frozen MNCs in medium. Frozen MNCs should be rapidly resuscitated at 37 °C before washing and washed once with medium;
3. Inoculate CB-MNCs resuspended in culture medium into T25 flasks at a seeding density of 3×10 5 cells/cm 2 ;
4. Place the cells in a 37 ° C 5% CO 2 incubator;
5. Change the medium every 7 days until the adherent fibroblast-like cells reach confluence;
6. Once confluence is reached, treat with 0.05% trypsin/EDTA for 5 min, resuspend the cells, and then re-inoculate the cells at 1 × 10 5 /bo;
7. After reaching the cell confluence, dilute with the medium 1:5 and then inoculate the culture;
